Also known as Schisandrol B, this bioactive lignan monomer is purified from Schisandra chinensis, well‑recognized for its prominent hepatoprotective properties. It exerts biological effects mainly by modulating the pregnane X receptor (PXR), a vital regulator responsible for bile acid homeostasis.
This compound can markedly boost the expression levels of multiple PXR‑downstream target genes associated with bile acid metabolism. In mouse liver tissue, it up‑regulates Cyp3a11, Ugt1a1, Oatp2 and Mrp3; meanwhile, in HEK293T cell lines, it elevates the expression of human CYP3A4, UGT1A1 and OATP2, thereby optimizing hepatic bile acid clearance and detoxification function.
Pharmacological studies have demonstrated its protective effect against liver injury triggered by lithocholic acid. It is capable of alleviating lithocholic‑acid‑provoked hepatic necrosis as well as intrahepatic cholestasis in wild‑type mice, whereas such protective effects disappear in Pxr‑deficient mouse models. When dosed at 100 mg/kg twice daily, it can effectively reduce mortality rates in cholestatic‑disease mouse models. Beyond anti‑cholestasis benefits, Gomisin A also facilitates liver tissue repair and regeneration after partial hepatectomy surgery.
